What keyboard are you using?

Generally:
-- plug the keyboard into a USB port of your computer
-- start up SampleTank
-- in Settings / Audio MIDI set up, select your keyboard and audio interface

Details are in the SampleTank User Manual - chapter 2.

I think I figured it out (for myself at least). From what I can tell, you have to download the standalone version as well as the vst version of your choice. I started off by only installing the vst3 to use inside my daw and and the same issue as above, clicking on the settings icon (gear looking thing) did not give me any audio or midi options. It wasn't until I reinstalled it with vst3 and standalone, went into the standalone version and clicked on the settings there was there the option of midi input. Once done there it should automatically work as a plug-in. Hope this helps someone.

The audio / MIDI settings in ST4's Preferences (or any other IK Multimedia software product) are only available in the stand-alone edition.

In the plug-ins, the audio and MIDI settings are handled by the DAW. Consult its user manual. Then you need to connect the DAW's MIDI input and audio output to the plug-in.

See 2.1.1 and 2.3 in the St4 User Manual.
